NanoExpo has developed a quick and easy system for the accurate titration of baculovirus stocks based on probe-based quantitative PCR (qPCR). In this approach, a freshly amplified baculovirus inoculum is added to a qPCR reaction mixture and then amplified. Following amplification, the quantity of virus particles is measured against titrated standards using the plaque assay. Our experience has shown that this method is as accurate as the plaque assay, but it is not recommended for virus stocks older than three months. Due to the aggregation and decomposition of the virus particles, old viral stocks produce erroneous results. Therefore, we advise using QuickCTtiter strictly with fresh virus stocks.
